Magnetic Bead DNA Extraction Kit Specially Used for Marine Organism
1, Lysis: Use hand drill to get 0.2g clean cancellous substance, mix with 0.3ml 1M DDT, grind them into thin mud, then take 0.1g ( about green bean size ) out and put it into a 1.5ml EP tube. Add PK 20µL and 300µL lysis buffer. Vibrate evenly and incubate it at 65°C for 1 hour. After taking it out, use a transferpettor ( its mouth wrapped with double layer thin silk net) to get 200µL supernatant and put it into another new EP. This can also be done after 10min centrifugation. For tissues like muscle and internal organs, they should be cut into small particles first, then do things as above stated.
2, Bind: add well-stirred 200µL magnetic bead liquid into the EP tube, suck it 8 times by using a transferpettor ( suck to 500µL level ) to mix up well, leave it at room temperature 25 °C for 6 min, slightly shake the tube several time every 3 min.
3, Wash: Insert the EP tube into the magnetic rack to allow all the magnetic beads to be absorbed onto the wall of the tube, then fully suck the unnecessary liquid( beads shall not be sucked ).Take the EP tube off the rack, add 600µL wash buffer, and suck it 8 times by using a transferpettor ( suck to 700µL level ) to mix up well, leave it there for 30sec. Then reput it onto the magnetic rack to allow all the magnetic beads to be absorbed onto the wall of the tube, then fully suck the unnecessary liquid. Repeat process 3 for one more time. Open the top to allow the beads to dry for 3min at room temperature.
4, Elute: Get the EP tube, add 100µL elute buffer, and suck it 8 times by using a transferpettor ( suck to150µL level ) to mix up well. Then put the tube into a constant temperature ( 65°C ) water tank to incubate 10 minutes, slightly shake the tube several times every 3 min. After 10 min, get the EP tube out, do the magnetic separation immediately, transfer the supernatant carefully into another new EP tube in order to do further experiments!
