1. Introduction Avian influenza is caused by Orthomyxoviridae type A family specific virus. There are 3 types of Influenza virus: A, B, C, as far as we know, only type A influenza virus can infect Poultry. Avian influenza virus (AIV) antigen rapid test card is based on rapid immunochromatographic detection technology, to detect avian influenza virus antigen in birds’ tracheal and cloacal swabs (or feces). The sample to be tested is added to the sample hole, it will move along with colloidal gold-labeled anti-AIV Monoclonal antibody on the chromatography membrane. If there is AIV antigen in the sample, the antigen will combine with the antibody on test line, appear red wine color. If there is not AIV antigen in the sample, no color appears. This rapid test is based on double antibody sandwich, using colloidal gold immunochromatographic method, to detect avian influenza virus. 2. Contents 1 AIV Ag test card 50 pieces 2 Swab 50 pieces 3 Dropper 50 pieces 4 Sample buffer 50 bottles 5 Disposable gloves 10 pairs 6 Manual 1 piece 3. Test procedure 1) Use swab to collect sample from cloaca (recommended) or tracheal, insert the swab into cloaca or tracheal and wipe, rotate in the inner wall repeatedly. For young birds, it can use swab to collect fresh feces sample directly. 2) Insert the swab into the tube with sample buffer immediately, rotate forcefully for at least 10 times in the tube wall. Mix the solution evenly, make the sample been dissloved in the buffer fully. Press swab on the tube wall above the solution level, so the solution can be extruded at last, discard the swab. 3) If there are big particles of feces, be static for 1-2 min, make big particles precipitation. 4) Take out test card from sealed bag. Put it on flat surface, use dropper to draw upper bright liquid, drop 4 drops slowly, one by one, to the sample hole with “S” mark. 5) Read the result in 10-15 minutes, the result is invalid after 15 minutes. 4. Results judgement 1) Negative: only control line is seen wine red;2) Positive: control line and test line are both seen wine red;3) Invalidation: control line isn’t seen wine red.
